The deletion of a single nucleotide that code a small sequence of a gene results, affect the release of insulin outside the cells because the single sequence is responsible for carrying the insulin outside the cells . The cells of pancreas are producing the  insulin normally but due to mutation in the singal sequence , due to which it can't excrete out of the cells and accumulate into the cytoplasm.
